# Parabilis Raised $670M to Drug the Target Oncology Couldn't Touch

$670 million. That was the gross haul when Parabilis Medicines opened on Nasdaq on June 10 — the largest US biotech IPO of 2026\. The underwriters had marketed the deal at $17 to $19 a share. The book priced at $20, and the offer grew from 25 million shares to 33.3 million.

The first session closed 58% higher. By late August the stock traded 96% above its offer price, while much of the year's biotech cohort gave its gains back. A record listing, held by a company most investors had never heard of six months earlier.

## Fog Pharma, then everything else

The company started in 2016 as Fog Pharmaceuticals, a Cambridge, Massachusetts lab built around one stubborn claim: the proteins inside a cell had been written off as undruggable because no conventional small molecule or antibody could reach them. Greg Verdine, a chemist who had already built and sold several protein-focused companies, founded it to prove otherwise.

## What a Helicon actually is

A Helicon is a stabilised alpha-helical peptide — a short chain of amino acids locked into a spring shape that crosses the cell membrane and presses flat against an intracellular protein. Parabilis calls them intracellular antibodies. The analogy holds on the mechanism. An antibody binds its target outside the cell. A Helicon binds one inside it.

Zolucatetide, the lead program, blocks the interaction between β-catenin and T-cell factor (TCF), the molecular switch that keeps the Wnt signalling pathway running. Most sporadic desmoid tumours carry a mutation in the CTNNB1 gene that removes the brake on that switch, so cells keep dividing when they should stop. Three decades of drug programs failed to touch the interaction directly. Zolucatetide is the first to try.

## The deal that priced the platform

Regeneron signed its collaboration on May 18 — three weeks before the listing. The terms read like a valuation: $125 million upfront in cash and equity, up to $2.2 billion across development, regulatory and commercial milestones, and tiered royalties on anything that reaches the market.

What Regeneron bought is an Antibody-Helicon Conjugate (AHC) — an antibody that carries a Helicon payload to a chosen cell type, then releases a peptide that hits a protein inside that cell. The buyer had just absorbed setbacks in its own cancer portfolio. Its existing antibody stack could not reach those targets. Here, the limitation became the purchase.

## The cash and the clock

The company ended the second quarter with $1.1 billion in cash, equivalents and marketable securities. Management says that funds operations into 2030\. The listing proceeds carry a specific map: $150 million for the desmoid trials, $120 million to widen zolucatetide into other indications, and $190 million to push earlier Helicon programs toward the clinic.

The Phase 3 registrational trial in desmoid tumours is scheduled to begin in the first half of 2027\. One readout carries the platform.

## Where it could go wrong

Desmoid tumours are rare. A clean Phase 3 win still lands in a small commercial pool, and the wider case leans on FAP and other Wnt-driven solid tumours that have not produced human data yet. One asset carries concentration risk that a portfolio company spreads across several bets.

Competition in engineered peptide platforms is no longer thin. Bicycle Therapeutics is running its own clinical bicyclic peptide programs, and Generate Biomedicines listed in February with a $400 million IPO on machine-designed proteins. Both compete for the same investor thesis: that engineered peptides can reach what antibodies cannot.

The listing window that made the IPO possible has narrowed since. BioBucks counted 21 US biotech IPOs in 2026, then logged a fourth straight week without a new pricing by late August. It rode the top of that wave. A company that lists at the peak inherits the downside of the cycle that follows.

## What the number really measures

Strip the ticker away and the story turns on modality. Peptide therapeutics sat at the edge of oncology for years — good in metabolism, hard to aim at intracellular targets. Helicons add cell penetration and a shape built to bind flat protein surfaces. Regeneron paid before retail money arrived, which is the cleaner signal.

Zolucatetide is the first test of whether that work clears a Phase 3\. If it does, $670 million stops looking like a record and starts looking like a platform price.
